چکیده

Article history: Received on: 19/03/2017 Accepted on: 12/05/2017 Available online: 30/06/2017 Cyclosporine A (CsA) is a widely used immunosuppressant in transplantation medicine. However, it has oxidative stress-induced side effects, including nephrotoxicity, hypertension, dyslipidemia, and glucose intolerance; particularly, post-transplant diabetes mellitus, accompanied by alterations of pancreatic islet cells, has been reported as a major adverse event. Recent research indicates that omega-3 polyunsaturated fatty acids, such as eicosapentaenoic acid (EPA), have antioxidant and anti-inflammatory activities. However, protective effects of EPA against CsA-induced alterations of pancreatic islet cells and hyperglycemia have not been examined in long-term administration of CsA in rats. The aim of this study was to determine whether EPA can abrogate these negative effects by assessing pancreatic islet cell dysfunction, vacuolation, and hyperglycemia in rats continuously treated with CsA. These pathologies were significantly improved by concomitant administration of EPA. Importantly, EPA did not affect the immunosuppressive effects of CsA, as evidenced by nuclear translocation of nuclear factor of activated T-cells in pancreatic tissue in response to lipopolysaccharide stimulation. Thus, EPA can be used to prevent CsA-induced oxidative stress and/or inflammation in pancreatic islets. Our data also suggest that EPA can be used in conjunction with CsA to mitigate its harmful side effects while preserving its immunosuppressant properties in transplant patients.

Salvianolic acid B improves insulin secretion from interleukin 1β-treated rat pancreatic islets: The role of PI3K-Akt signaling

Background and Objective: Oxidative stress induced by proinflammatory cytokines such as IL-1β plays a major role in β-cell destruction in diabetes type 1. Salvianolic acid B (Sal B) is a polyphenolic compound with antioxidant and protective effects. Thus, objective of this study was to assess the protection exerted by Sal B on isolated rat islets exposed to IL-1β and to investigate an underlyin...

Oral Administration of Ginseng Ameliorates Cyclosporine-Induced Pancreatic Injury in an Experimental Mouse Model

BACKGROUND This study was performed to investigate whether ginseng has a protective effect in an experimental mouse model of cyclosporine-induced pancreatic injury. METHODS Mice were treated with cyclosporine (30 mg/kg/day, subcutaneously) and Korean red ginseng extract (0.2 or 0.4 g/kg/day, oral gavage) for 4 weeks while on a 0.01% salt diet. The effect of ginseng on cyclosporine-induced pan...

Correlation between lipid-lowering and bone-protective effects of eicosapentaenoic acid in rats with steroid-induced bone loss

This study was conducted to investigate the correlation between lipid-lowering and bone-protective effects of eicosapentaenoic acid on steroid-induced bonechanges in rats. Twenty-one male 10-week-old Wistar rats were divided into 3 groups (n = 7 each) and treated with 0.9% NaCl SC (group 1) or methylprednisolone 7 mg/kg SC once a week (group 2) or methylprednisolone 7 mg/kg SC once a week + eic...

Omega-3 fatty acids prevent pressure overload-induced cardiac fibrosis through activation of cyclic GMP/protein kinase G signaling in cardiac fibroblasts.

BACKGROUND Omega-3 polyunsaturated fatty acids (eicosapentaenoic acid and docosahexaenoic acid) from fish oil ameliorate cardiovascular diseases. However, little is known about the effects of ω-3 polyunsaturated fatty acids on cardiac fibrosis, a major cause of diastolic dysfunction and heart failure. The present study assessed the effects of ω-3 polyunsaturated fatty acids on cardiac fibrosis....

Renoprotective Effects of Naringenin and Olive Oil against Cyclosporine- Induced Nephrotoxicity in Rats

Background: Prolonged use of cyclosporine A for prevention of allograft rejection is associated with nephrotoxicity development. Naringenin and olive oil are beneficial dietary antioxidants with potential renoprotective properties. This study aimed to evaluate the therapeutic effect of naringenin and olive oil in alleviating cyclosporine induced nephrotoxicity in rats by the assessment of renal...
